         染色体端粒是染色体末端的重复核苷酸元件,保护染色体免受降解和遗传信息丢失。正常的二倍体细胞在每个细胞周期中都会失去端粒。因此,端粒长度随着时间的推移而减少,并可能预测寿命。端粒缩短对健康状况有负面影响,并与许多健康问题有关,包括衰老和癌症。准确和一致的端粒长度定量在细胞生物学的许多方面都很重要,如染色体不稳定性、DNA修复、衰老、凋亡、细胞功能障碍和肿瘤发生。


         ScienCell的绝对人类端粒长度定量qPCR检测试剂盒(AHTLQ)设计用于直接测量人类细胞群的平均端粒长度。端粒引物组识别和扩增端粒序列。单拷贝参考(single copy reference, SCR)引物集识别并扩增人类17号染色体上一个100 bp长的区域,作为数据归一化的参考。已知端粒长度的参考基因组DNA样本作为计算目标样本端粒长度的参考。精心设计的引物确保:(1)高效可靠的定量;(ii)无非特异性扩增。每组引物均经熔体曲线分析和凝胶电泳qPCR验证扩增特异性,模板连续稀释验证扩增效率。2X GoldNStart TaqGreen qPCR主混合物(Cat #MB6018a-1)是一种SYBR®绿色染料基qPCR主混合物,具有“热启动”特性。它包含SYBR®绿色,dNTPs, Taq DNA聚合酶,惰性金色负载指示器在单管中。通过ScienCell独特的化学修饰Taq DNA聚合酶实现的“热启动”特性,最大限度地抑制了引物二聚体的形成。先进的缓冲配方提供了卓越的专一性和效率与宽线性动态范围。惰性金色上样指示器可以更好地可视化和跟踪qPCR板或管中的样品上样。
